Abstract. Grass cover along a grazing intensity gradient in Patagonia decreases, whereas bare soil and shrub cover increases. Our objective was to study the effect of a change in the dominant plant functional type on soil water balance, primary production, herbivore biomass, roughness, and albedo. Using a soil water balance model, we found increases in evaporation and deep drainage, and a decrease in total transpiration along the grazing intensity gradient. Above-ground primary production, estimated from transpiration, decreased along the grazing intensity gradient because shrubs did not fully compensate for the decrease in grass production. Using a statistical model, we calculated herbivore biomass from estimates of above-ground primary production. Estimated herbivore biomass was lowest in the shrub-dominated extreme of the grazing gradient. Roughness increased from the grass-dominated to the shrub-dominated community. Albedo had a maximum at an intermediate position along the gradient. Our results suggest that changes in plant functional type composition, independent of changes in biomass, affect ecosystem functioning and the exchange of energy and material with the atmosphere. Grasses and shrubs proved to be appropriate plant functional types to link structure and function of ecosystems.  相似文献

Abstract. A vegetation map at scale 1:5 million is presented. * * Attached on the inside of this issue's back cover.
It covers Bangladesh, Burma (Myanmar), India, Cambodia, Laos, Thailand, Vietnam and Sri Lanka and fills a conspicuous gap in the cartography of tropical vegetation, following the publication of vegetation maps of South America, Africa and Malaysia. For conformity, it is presented as one sheet at a scale of 1:5 million. It uses the basic map of the American Geographical Society (1942; bipolar oblique conformal projection) which forms the base for FAO's Soil map of the world. Basic information was obtained from many published maps, unpublished observations and satellite data. The limits of the main vegetation types have been updated with a complete set of Landsat MSS images (369 scenes) with a mosaic of Landsat TM data for 1991 and with recent forest maps from Asia. Nine main vegetation units, which are groups of forest formations, have been identified and mapped, including woodlands, thickets and wooded savannas. Agricultural land has been shown in a uniform pale green colour in order to clearly express the extent of human impacts on woody vegetation. In spite of the necessary oversimplification of the ground data, this map is probably the most explicit expression of the remaining forest stands and of the regression of natural vegetation in the region. It can be considered as a benchmark for future monitoring of tropical vegetation.  相似文献

Immunochemical probes are widely used to identih different species and to quantify and understand the role that different antigens play within cells. We optimized a single-cell immunofluorescent assay for the carbon fixation enzyme ribulose-1,5-bisphosphate carboxylase (Rubisco) in order to quantify the enzyme by flow cytometry in phytoplankton cells. The criteria for optimization of the immunofluorescent assay for Rubisco in single cells included maximization of Rubisco immunogenicity, minimization of Rubisco diffusion out of the cells, minimization of cell breakage, and maximization of the cell labeling. Several fixatives (cross-linkers and denaturing) and permeabilizing agents were tested on 26 species of phytoplankton. The only fixative / permeabilizing agent that fulfilled the criteria established for the assay was 96% ethanol. Phytoplankton cells collected from the field needed further treatment with a strong oxidant to permeabilize ethanolfixed cells and thus allow the antibody probe to access the Rubisco antigen. This study should have a general applicability to the study of other soluble photosynthetic antigens in single phytoplankton cells.  相似文献

In order to better understand the biochemical mechanisms of DNA metabolism in chloroplasts, repair of UV induced plastome damage in vivo was determined by exposure of soybean suspension cells to UV light and subsequent quantitation of the damage remaining in nuclear and chloroplast encoded genes with time by quantitative polymerase chain reaction (QPCR). The kinetics of damage rapir in the nuclear rbcS gene suggest that photoreactivation and dark mechanisms are active, while for the plastome encoded psbA gene only a light-dependent repair process was detected which is considerably slower than would be expected for photolyase-mediated photoreactivation.  相似文献

Summary— Multidrug-resistant (MDR) variants of a human osteosarcoma cell line (U-2 OS) have been recently obtained by continuous exposure to doxorubicin (DX). The growth and phenotypic characteristics of these cell lines have been demonstrated to be related to the level of expression of P-glycoprotein. In this work, the morphological changes associated with MDR have been evaluated by quantitative image analysis and transmission electron microscopy. Resistant cells present morphological changes with respect to sensitive cells at both cytoplasmic and nuclear level. Some of these changes appear to be related to the degree of resistance but not to the direct presence of DX, since deprived cells maintain some modified characters, while others are partly lost. These findings suggest that DX exposure affects cell metabolism causing progressive changes of the cell morphotype.  相似文献

This study aims to develop an integrated methodology applying Remote Sensing and Geographic Information Systems (GIS) techniques for the assessment of the ecological status of mangrove forests. The study area is located at Phangnga Bay, Thailand. Various commonly available remote sensing data are evaluated for mangrove vegetation mapping. The satellite sensors used are covering the visible and infra-red (VIR) spectrum up to the microwave region of the electromagnetic spectrum. This study provides recommendations regarding the selection of a single sensor approach or sensor combination that fulfills a minimum requirement for practical mangrove mapping and inventory purposes (e.g. mangrove and non-mangrove areas, varying stocking density, dominant species composition and impact of human activities). We hypothesize that the evolution of an ecologically important character, the host associations of specialized phytophagous insects, has been influenced by limitations on genetic variation. Using as a historical framework a phylogenetic reconstruction of the history of host associations in the beetle genus Ophraella (Chrysomelidae), we have employed quantitative-genetic methods to screen four species for genetic variation in larval survival, oviposition (in one species only), and feeding responses to their congeners' host plants, in the Asteraceae. We here report results of studies of one species and evaluate the results from all four. Analysis of half-sib/full-sib families and of progenies of wild females of O. notulata, a specialist on Iva (Ambrosiinae), provided evidence of genetic variation in larval consumption of five of six test plants and in adult consumption of four of six. Larval mortality was complete on five plants; only on Ambrosia, a close relative of the natural host, was there appreciable, and genetically variable, survival. Oviposition on Ambrosia showed marginally significant evidence of genetic variation; a more distantly related plant elicited no oviposition at all. In compiling results from four Ophraella species, reported in this and two other papers, we found no evidence of genetic variation in 18 of 39 tests of feeding responses and 14 of 16 tests of larval survival on congeners' hosts. This result is consistent with the hypothesis that absence or paucity of genetic variation may constrain or at least bias the evolution of host associations. The lower incidence of genetic variation in survival than in feeding behavior may imply, according to recent models, that avoidance is a more common evolutionary response to novel plants than adaptation. The usually great disparity between mean performance on congeners' hosts and the species' natural hosts, and an almost complete lack of evidence for negative genetic correlations, argue against the likelihood that speciation has occurred by sympatric host shift. The presence versus apparent absence of genetic variation in consumption was correlated with the propinquity of relationship between the beetle species tested and the species that normally feeds on the test plant, suggesting that the history of host shifts in Ophraella has been guided in part by restrictions on genetic variation. It was also correlated with the propinquity of relationship between a test plant and the beetle's natural host. The contributions of plant relationships and insect relationships, themselves correlated in part, to the pattern of genetic variation, are not readily distinguishable, but together accord with phylogenetic evidence that these and other phytophagous insects adapt most readily to related plants. In this instance, therefore, the macroevolution of an ecologically important character appears to have been influenced by genetic constraints. We hypothesize that absence of the structural prerequisites for genetic variation in complex characters may affect genetic variation and the trajectory of evolution.  相似文献

Heterogeneous phenotypic correlations may be suggestive of underlying changes in genetic covariance among life-history, morphology, and behavioural traits, and their detection is therefore relevant to many biological studies. Two new statistical tests are proposed and their performances compared with existing methods. Of all tests considered, the existing approximate test of homogeneity of product-moment correlations provides the greatest power to detect heterogeneous correlations, when based on Hotelling's z*-transformation. The use of this transformation and test is recommended under conditions of bivariate normality. A new distribution-free randomisation test of homogeneity of Spearman's rank correlations is described and recommended for use when the bivariate samples are taken from populations with non-normal or unknown distributions. An alternative randomisation test of homogeneity of product-moment correlations is shown to be a useful compromise between the approximate tests and the randomisation tests on Spearman's rank correlations: it is not as sensitive to departures from normality as the approximate tests, but has greater power than the rank correlation test. An example is provided that shows how choice of test will have a considerable influence on the conclusions of a particular study. A greenhouse assay to screen sunflower for resistance to Alternaria helianthi is described. A comparison of conditions led to the following standard conditions being recommended. The first or second pair of leaves of seedling plants at the V8 growth stage are inoculated using inoculum grown on sunflower leaf extract agar for 5–10 days at an inoculum density of 1–2 spores cm2 of leaf tissue. A 48 h dew period should be applied to plants covered by a plastic tent. A dew period temperature of 26/26°C night/day and a post-dew period temperature relative to that experienced under local growing conditions should be applied. Lesions are measured 7 days after inoculation, and mean lesion size per plant is calculated. Mean lesion size of lines being tested is expressed as a proportion of the mean lesion size of a susceptible standard included in each screening experiment.  相似文献
